Nonoxynol-9 or N-9 or Nonoxinol 9 is a common spermicide used in vaginal contraceptive products like foams, gels, films, and sponges to prevent pregnancy by damaging sperm membrane. While available over the counter for over 60 years, it does not protect against STIs or HIV and can cause vaginal irritation, increasing infection risks if used frequently.

Nonoxynol 9 is an anhydrous liquid mixture consisting chiefly of monononylphenyl ethers of polyethylene glycols corresponding to the formula:
C9H19C6H4(OCH2CH2)nOH
in which the average value of n is 9. It contains NLT 90.0% and NMT 110.0% of Nonoxynol 9.
IDENTIFICATION
A. Infrared Absorption.
B. The retention time of the Sample solution corresponds to that of the Standard solution.
Assay: 90% to 100%
Free Ethylene Oxide: To pass the test by chromatography.
Limit of Dioxane: To pass the test.
Polyethylene Glycol: To pass the test.
Cloud Point: To pass the test. Acceptance criteria: 52C to 56C.
Fats and Fixed Oils, Acid Value: NMT 0.2
Water Determination: NMT 0.5%
Packaging and Storage: Preserve in tight containers.

Action and use: Spermatocide.
DEFINITION
α-(4-Nonylphenyl)-ω-hydroxynona(oxyethylene).
Mixture consisting mainly of monononylphenyl ethers of macrogols corresponding to the formula: C9H19C6H4-[OCH2-CH2]n-OH where the average value of is 9. It may contain free macrogols.
CHARACTERS
Appearance: Clear, colourless or light yellow, viscous liquid.
Solubility: Miscible with water, with ethanol (96 per cent) and with vegetable oils.
IDENTIFICATION
A. Infrared absorption spectrophotometry.
B. Cloud point.
TESTS
Acidity or alkalinity: Boil 1.0 g with 20 mL of carbon dioxide-free water for 1 min, with constant stirring. Cool and filter. To 10 mL of the filtrate, add 0.05 mL of bromothymol blue solution. Not more than 0.5 mL of 0.01 M hydrochloric acid or 0.01 M sodium hydroxide is required to change the colour of the indicator.
Hydroxyl value: 84 to 94.
Cloud point: 52C to 58C.
Ethylene oxide and dioxan: Maximum 1 ppm of ethylene oxide and maximum 10 ppm of dioxan.
Water: Maximum 0.5 per cent, determined on 2.00 g.
Total ash: Maximum 0.4 per cent, determined on 1.0 g.
Storage: In an airtight container.
